Description:The dose/response relationship for breathing rate and cellular responses were determined in English-short-hair-guinea-pigs exposed to cotton dust. Responses were measured 18 hours after a 6 hour exposure of guinea-pigs to mass concentrations of cotton dust ranging from 0.4 to 32mg/m3. Various cellular and mechanical responses of the guinea-pig lung exhibited different sensitivities to cotton dust exposure. The most dramatic response was the migration of leukocytes into the pulmonary air space. The data indicate that at 18 hours after exposure to concentrations of cotton dust above 5.5mg/m3, the increase in leukocyte yield began to decline. Similar relationships were noted for alveolar macrophages. A relationship was noted between the migration of leukocytes into the lung and the secretory activity of alveolar macrophages. Apparently alveolar macrophage activity depends on the mediators secreted from pulmonary leukocytes. Depletion of circulating leukocytes completely abolished the cotton dust induced activation of Zymosan stimulated superoxide anion release from alveolar macrophages. [Description provided by NIOSH]
