Description:In order to determine whether vitamin-C was an effective treatment for byssinosis, the effect of dietary ascorbic-acid on the pulmonary response of the guinea-pig animal model to cotton dust exposure was evaluated. The effects of deficiency, normal supplementation and mega supplementation of this vitamin on the response to the inhalation of cotton dust was investigated. Dietary deficiency of vitamin-C did not enhance and supplementation did not prevent the acute response to cotton dust exposure in the animal model of byssinosis. Vitamin-C deficient animals demonstrated a lower carbon- dioxide challenge breathing rate at 18 hours following exposure than vitamin-C supplemented animals. After exposure to cotton the magnitude of the influx of leukocytes into the airspaces was slightly increased with mega supplementation of vitamin-C. The activity of alveolar macrophages, measured by the ability to produce superoxide anion in response to Zymosan stimulation, was increased in cotton exposed guinea-pigs receiving mega supplemental doses of vitamin-C. Rather than being protective, vitamin-C appears to enhance the pulmonary responses of the guinea-pig model to cotton dust exposure. These findings are in contrast with reported results among humans.
