Description:Male Sprague-Dawley-rats were exposed to an antigen extract and antigen plus endotoxin in a study of the inhibition of interleukin-1 (IL-1) in an inflammatory reaction resulting from exposure to Aspergillus-umbrosus. Rats were exposed three times, 3 days apart to antigen extract at 100 microliter/100 grams body weight and endotoxin at 1 million endotoxin units/milliliter, or to antigen extract alone. Alveolar macrophages were obtained through lavage. Exposure to A-umbrosus increased IL-1 activity 12 times in alveolar macrophages in-vivo. Total cell counts showed that the exposure caused an inflammatory condition in the lungs because the number of macrophages in the lavage fluids of the exposed rats increased from could increase IL-1 activity more than exposure to A-umbrosus plus endotoxin. The authors conclude that A-umbrosus can cause an IL-1- mediated inflammatory reaction similar to endotoxin. A-umbrosus is one of the causative agents of hypersensitivity pneumonitis. The authors suggest that IL-1 may also be involved in the pathogenesis of hypersensitivity pneumonitis. [Description provided by NIOSH]
