Description:The composition and toxicity of a commercial leather conditioner that was responsible for a nationwide outbreak of respiratory illness in 1992 were studied. The outbreak of respiratory illness occurred with the use of a new formulation of an existing leather spray product. The chemical characteristics of the original and new spray formulations were studied and their toxic effects were assessed following inhalation exposure of guinea-pigs and Sprague- Dawley-rats for 2 hours. Both the original and new formulations contained propane (74986) and C7 to C8 alkanes as the major solvent components. The new formulation also contained traces of 2- butoxyethanol (111762), isomers of dipropylene-glycol-methyl-ether (34590948), and additional C10 to C12 alkanes as well as altered fluorohydrocarbon constituents. Exposure to aerosol from the new formulation resulted in the death of 33% or more of the animals, induced tachypnea, increased the number of erythrocytes and alveolar macrophages in bronchoalveolar lavage fluid, and induced pulmonary lesions characterized by alveolar hemorrhage, edema, and necrosis of type-I cells. The authors conclude that these studies confirm the toxicity of the new leather conditioner formulation and provide potential candidates for specific etiologic agents. [Description provided by NIOSH]
