Description:The cellular composition of lung lavage fluid was studied in dogs. The chemiluminescence of the cells was also measured. The lungs were lavaged three times with 600 milliliters (ml) of 0.16 molar sodium-chloride containing 1 International Unit/ml sodium-heparin. The washings were pooled and centrifuged. After centrifugation, the concentrated cell suspensions underwent centrifugal elutriation to separate the cells. The cells were counted and sized with a Coulter counter and examined by light and transmission electron microscopy. Chemiluminescence of the cells was measured by a liquid scintillation counter after activating the cells with zymosan. The cells in the lavage fluid were separated into three fractions by centrifugal elutriation. The fractions had mean cellular volumes peaking at 50 cubic microns (micron3) (fraction-I), 150 to 200micron3 (fraction-II), and 1000micron3 (fraction-III). Fraction- I was composed of cellular debris, lymphocytes, and red blood cells, fraction-II small granular cells, and fraction-III alveolar macrophages. Fraction-II and Fraction-III showed chemiluminescence. The maximum chemiluminescence of fraction-II was about five times greater than that of fraction-III. The authors conclude that dog lung lavage fluid consists of three fractions including alveolar macrophages. The unwanted cells can be separated from the alveolar macrophages by centrifugal elutriation. Since the levels of chemiluminescence differ between the two fractions, cells of fraction-II and fraction-III may have different functions. [Description provided by NIOSH]
