Description:Styrene-oxide (96093) (SO) was subjected to a tier II mutagenic test screening program. The screening included the following assays: unscheduled DNA synthesis (UDS) assay in human diploid fibroblasts with exposures of 3 hours duration and concentrations up to 50 micrograms per milliliter of culture medium; dominant lethal test in male rats with exposure to atmospheres containing 15 or 100 parts per million (ppm) SO styrene-oxide for 7 hours per day for 5 consecutive days; sperm abnormality test in male mice using the same conditions; cytogenetic test in male and female rat bone marrow cells using the same exposure conditions or a single exposure of 7 hours duration; and sex linked recessive lethal test in Drosophila- melanogaster with exposure of 100ppm for 150 minutes. There was no increase in UDS in cells treated with SO and the frequencies of chromosomal aberrations were not increased in the rat bone marrow cells. There was a small effect attributable to SO in the dominant lethal test on pregnancy frequency and on the total number of implantations. Sperm abnormality frequency was slightly increased in mice dosed with 100 parts per million SO, while sex linked recessive lethal mutation frequency was not increased. The author concludes that SO did not show genetic effects detectable in the experiments apart from the small increase in the frequency of sperm with abnormalities. [Description provided by NIOSH]
