Description:The use of in-vigro tests for detecting chemical mutagenicity is evaluated. Types of aberrations produced in cell culture systems are discussed, including DNA alkylation, arylation and acylation. Mutations arising from antireplication effects are considered. The influence of metabolic activation is reviewed and specific sites of action are described. Structure activity relationships and correlations between carcinogenicity and mutagenicity in Salmonella tests are discussed. The authors conclude that eukaryotic organisms should be tested in both in-vitro and in-vivo systems before mutagenicity hazards can be evaluated. [Description provided by NIOSH]
