Validations of time-resolved fluoroimmunoassays for urinary estrone 3-glucuronide and pregnanediol 3-glucuronide

Description:Competitive time resolved fluoroimmunoassays (FIAs) for measuring steroid glucuronides in urine were developed. The new procedure avoided the use of radioisotopic reagents. Urine samples for validation studies were collected from men and nonpregnant women. Mean recoveries for added urinary steroid glucuronides were 102.8+/- 2% for estrone-3-glucuronide (E13G) and 93.6+/-2.9% for pregnanediol- 3-glucuronide (Pd3G). Recovery was not dependent on the concentrations of added analyte. The limits of detection for the E13G and Pd3G FIAs were 0.98 nanograms/milliliter and 0.035 micrograms/milliliter, respectively. Urinary FIA measurements correlated well with radioimmunoassay (RIA) measurements in urine and serum. However, urinary Pd3G values measured by FIA were about 2.5 times greater than those measured by RIA. The luteal peak/follicular phase PdG ratio was 50% greater when measured by FIA compared to RIA. The authors conclude that the FIAs can be used to evaluate menstrual function in epidemiological, clinical and sports populations. [Description provided by NIOSH]
