Description:The profiles of volatile metabolites in serum samples obtained from normal individuals and diabetic patients with symmetrical distal polyneuropathy were assessed in relation to the production of endogenous neurotoxic diketones. Serum samples extracted with 2- chloropropane were analyzed by gas chromatography mess spectrometry using a transevaporator sampling procedure. Chromatograms from normal samples showed a consistent pattern of peaks. Varying chromatographic patterns were obtained from diabetic patients depending upon the degree of neuropathy and the drug treatment regimen. The chromatograms of patients receiving insulin were similar to the normal pattern, whereas the chromatograms of chronic diabetics receiving no therapy were showed significant differences from normal. No correlation was found between the severity of the neuropathy and the concentrations of specific ketones. The results were discussed in relation to the oxidation of 2-hexanone (591786) and 3-heptanone (106354) and their metabolites to form gamma diketones. [Description provided by NIOSH]
