Synergistic inactivation of plasma alpha1-proteinase inhibitor by aldehydes of cigarette smoke with styrene oxide and 1,2-dichloroethane.

Description:The interaction of aldehydes of cigarette smoke with styrene-oxide (96093) and 1,2-dichloroethane (107062) (DCE), and their effect on plasma alpha1-proteinase inhibitor (a1PI) were studied in-vitro. Solutions of purified a1PI were incubated with styrene-oxide, pyruvic-aldehyde (78988), acrolein (107028), or DCE alone or in combination. The effects on elastase inhibitory capacity (EIC) and trypsin inhibitory capacity (TIC) were determined. Styrene-oxide alone caused a concentration related decrease in EIC and TIC, which was enhanced when acrolein was added. Acrolein by itself had no effect. Styrene-oxide and pyruvic-aldehyde caused a similar enhancement of TIC and EIC inhibition. Styrene-oxide plus DCE and DCE plus pyruvic-aldehyde caused a synergistic inhibition of EIC and TIC. The enhanced effect was similar to that observed with styrene- oxide. Adding DCE or styrene-oxide to acrolein resulted in enhanced decreases in EIC and TIC. Adding acrolein or pyruvic-aldehyde to DCE resulted in a greater inactivation of EIC and TIC than with DCE alone; however, the effect was not as pronounced as with the other combinations. The authors conclude that a1PI can undergo synergistic inactivation by the chemicals and constituents of cigarette smoke. If a1PI is used as an index of chemical exposure, information on the smoking habits of the exposed population must be obtained. Since inactivation of a1PI is thought to be a key factor in the development of pulmonary emphysema, smokers working in the chemical industry may be at elevated risk of developing emphysema.
