Description:The mutagenicity of cyclopiazonic-acid (18172333) (CPA) with aflatoxin-B1 (1162658) (AFB1) was investigated by the Ames test using Salmonella-typhimurium TA-98 and TA-100. Mutagenicity was tested by the Ames histidine reversion assay with or without using S9 mixture for metabolic activation. Test concentrations ranged from 0.01 to 1.0 picomoles for CPA and from 0.03 to 30.0 picomoles for AFB1. A possible correlation between concentrations and the number of revertants per plate existed with metabolic activation for TA-98 and TA-100. No such correlation in either strain was evident when S9 mixture was not used in the test. Both CPA and AFB1 produced significant mutagenicity singly and in combination when studied in factorial experiments with TA-98. The mutagenic activity of the two compounds when tested together was additive rather than synergistic. The authors conclude that CPA is commonly produced by organisms such as Aspergillus-flavus and its presence in contaminated grain contributes to the genotoxic hazards of such grain.
