Description:The mutagenicity of 147 industrial chemicals and structurally related compounds was studied in-vitro. The Ames Salmonella- typhimurium microsome plate incorporation test was used in the assay system with tester strains TA1535, TA1537, TA98, and TA100. Five different concentrations of each chemical were tested with and without S-9 prepared from the liver of Sprague-Dawley-rats pretreated with Aroclor-1254 (11097691). Of the 147 compounds tested, 120 were not mutagenic to any of the testers, either with or without metabolic activation. Metabolic activation was required for the mutagenic activity of 1,2,3-trichloropropane (96184), dimethoxyethylphthalate (131113), 2,4-dimethylaniline (95681), and 2,6-dinitrotoluene (606202). Twenty-three compounds were directly mutagenic to one or more testers. The direct mutagens included nitromethane (75525), 2-nitropropane (79469), methyl-bromide (74839), methylene-chloride (75092), monochloroethane (75003), 2,3- dinitrotoluene (602017), 2,5-dinitrotoluene (619158), 2,4,5- trinitrotoluene (610253), p-dinitrobenzene (100254), and m- dinitrobenzene (99650).
