Description:The genetic activity of 5-nitroimidazole derivatives was studied. Escherichia-coli (E-coli), Saccharomyces-cerevisiae (S-cerevisiae), Neurospora-crassa (N-crassa), and Drosophila-melanogaster were exposed to metronidazole (443481) and three other 5-nitroimidazole derivatives (compounds 2, 3, 4). Forward and back mutations in the E- coli, forward mutations in N-crassa, mitotic gene conversion in S- cerevisiae, and sex linked recessive lethals in Drosophila were determined. Metronidazole was moderately mutagenic and compounds 2 and 3 were strongly mutagenic in all four test systems. The mutagenicity of 2 and 3 in E-coli was evident even at very low concentrations. Compound 2 had the strongest effect in Drosophila. Compound 4 was nonmutagenic in all tests. The authors conclude that the mutagenic activity of 5-nitroimidazoles differ depending on the test strain.
