Behavioral effects of acrylamide in the mouse.

Description:The effects of acrylamide (79061) on a test of appetite behavior, milk licking, was compared with standard measures of toxicity using both time effect and dose response functions. In addition, the suitability of using the mouse to evaluate behavioral toxicity and to characterize the behavioral effects of acrylamide was determined. Two strains of mice, CD-1-mice and C57BL/6J-mice, were injected intraperitoneally five times weekly with either saline, 20, 60 or 100 milligrams per kilogram (mg/kg) of acrylamide. Body weight loss, mortality, hindlimb grip strength, and locomotor activity were evaluated in parallel with milk licking. After 5 weeks of dosing with 20mg/kg, decreased hindlimb grip strength was observed but there was no effect on milk licking or body weight. Within three weeks of administration of the 60mg/kg dose, slight loss of body weight was evident as was neuropathy and a 50 percent mortality rate. In contrast, by day two of treatment with this dose there was a highly significant increase in episodic milk licking which persisted throughout the dosing period. Administration of 100mg/kg subchronically, but not acutely, resulted in weight loss and severe neuropathy within 3 days and a 100 percent mortality rate within 2 weeks. The authors conclude that compared to standard measures of toxicity, milk licking proved to be a more sensitive indicator of acrylamide toxicity, increases in milk licking were evident even in severely intoxicated mice and preceded other signs of toxicity, and that the magnitude of the daily dose rather than the cumulative dose is a better predictor of the onset of various toxic effects. They suggest that the mouse is a valid animal model for studies in behavioral toxicology. [Description provided by NIOSH]
