Description:Thin layer chromatographic (TLC) procedures were developed to determine toxins in airborne corn dust samples. Ground corn and airborne dust samples were collected and prepared for TLC analysis. Analysis included chloroform extraction of dust samples, purification of extracts on a silica gel column, and measurement of aflatoxin (1402682) by either one or two dimensional TLC. The solvent for one dimensional TLC was chloroform, acetone and water, while the solvents for two dimensional TLC were ether, methanol, and water for the first direction and chloroform, acetone, and water for the second direction. Alternatively, the two dimensional method used for the first direction chloroform, acetone, and water, and for the second direction toluene (108883) ethyl acetate (141786), and formic acid (64186). The mean aflatoxin-B1 ground corn. The limits of detection was about 9ng per gram for 0.1 gram samples. [Description provided by NIOSH]
