Description:The genotoxic and mutagenic effects of halothane (151677) and its metabolites were studied. Standard mutagenicity assays using Salmonella-typhimurium were performed for halothane, 2-chloro-1,1- difluoroethylene (359104), and 2-bromo-1,1-difluoroethylene. Bacillus-subtilis strains H17 and M45 were grown and treated with test compounds. Toxicities and DNA interactions were assessed. Mutagenicity assays were also performed using polymerase deficient Escherichia-coli. Halothane and its metabolites were nonmutagenic in Salmonella, however, conjugates of these compounds were weakly mutagenic. In Bacillus-subtilis, halothane metabolites inhibited growth of the M45 strain which is deficient in DNA repair ability. There was no differential toxicity found in tests for DNA modifying activity in Escherichia-coli. The authors conclude that there is a possible relationship between halothane metabolism and reported toxic effects associated with occupational anesthetic exposures. [Description provided by NIOSH]
