Description:A technique for preparing a viable population of Schwann cells based on the phenomenon of Wallerian degeneration was demonstrated. The sciatic nerves of albino-rabbits were exposed, crushed, and cut. The nerve stumps were then sewn into adjacent muscle surface to impede regenerative processes. Distal nerve stumps containing the proliferated Schwann cells were removed 5 to 12 weeks later, and morphologically studied by light and electron microscopy. Schwann cells formed longitudinal columns separated by endoneurial connective tissue. The authors conclude that this technique has the following advantages: simplicity and accessibility, large tissue yield, applicability to different species for comparative study, prolonged in-vivo viability and usefulness for analysis of axonal regulation of Schwann cell function and determination of the role of Schwann cell alteration in peripheral nerve reaction.
