Description:Optic nerve sections were grafted between the cut ends of the peroneal nerve of one limb, or alongside the intact peroneal nerve in the opposite limb as control, in 45 rats to investigate peripheral axon stimulation of oligodendrocyte myelination. The grafts and control transplants were examined by light and electron microscopy 4 to 28 weeks later. Control transplants were viable at all times. At 8 weeks, but not at 20 weeks, central nervous system (CNS) type myelin was found around oligodendrocyte cell bodies and circular astrocytic processes in proximity to the myelinated oligodendrocytes. The portion of the peroneal and optic graft originating from the proximal stump of the peroneal contained preserved and regenerated peripheral nervous system (PNS) myelinated fibers, and regenerated myelinated and unmyelinated fibers were found in the distal portion of the stump. PNS fibers were regenerated around and outside the original graft, and at 20 weeks, a few myelinated fibers, both PNS and CNS, surrounded by proliferated astrocytes, were located within the boundary of the original graft. The authors conclude that peripheral and central axons can each instruct both Schwann cells and oligodendrocytes to produce myelin, and the type of myelin they manufacture is cell specific and unrelated to the location of the myelinating cell. The authors suggest the existence of a similarity in the axon surface recognition properties of Schwann cells and astrocytes and that the astrocytes play an important role in establishing and maintaining the central and peripheral divisions of the nervous system.
