Description:The Tier II mutagenic screening of 1,1,2,2-tetrachloroethane (79345) (TCE) was conducted. The unscheduled DNA synthesis (UDS) assay was conducted with human embryonic intestinal cells, rat liver S9, and 1.3 to 15,869 micrograms per milliliter TCE. A cytogenetic analysis of rat bone marrow cells was performed after in-vivo exposure to 5 or 50 parts per million (ppm) TCE for 7 hours per day for 1 or 5 days. Dominant lethal tests with male rats and sperm abnormality tests with male mice were performed after in-vivo exposure to 5 or 50ppm TCE for 7 hours per day for 5 days. The sex linked recessive lethal test was performed using Drosophila-melanogaster after a 40 minute exposure to 50ppm or a 7 hour exposure to 5ppm. TCE produced no increase in UDS and no signs of toxicity in mice or rats at the test concentrations used. The only increase in rat bone marrow cell DNA aberrations was observed in females exposed to a single dose of 50ppm TCE. No effects due to TCE exposure were seen in the dominant lethal assay, the frequency of mouse sperm with hook abnormalities was increased slightly by exposure to 50ppm, and the sex linked recessive lethal mutation frequency was not increased. The author concludes that TCE was not tested to the limits of these assays and that they should be repeated.
