Description:An investigation was conducted using rats to determine the effects of repeated orally administered diglyme (111966) on the pathology and lactate-dehydrogenase-X (LDH-X) levels of the testis, the primary site of toxicity of certain other glycol ethers. Daily administration of diglyme at 5.1 millimole/kilogram body weight to male Sprague-Dawley-rats resulted in reduced body weight gain and reduced weights of certain organs. Significant reductions in the average body weight gain of the rats occurred after 18 consecutive daily doses of diglyme. The testes weights revealed reductions after as few as ten doses. The testes, epididymides, and thymus glands of rats given 20 doses of diglyme demonstrated significant decreases in relative weights. No such change was noted for brains. Some degenerative changes were noted on histopathological examination of the testes after 20 consecutive days of dosing. Mild degenerative changes were noted in the germinal cells after as few as eight doses. The early microscopic changes included loss of primary and secondary spermatocytes, loss of sperm bundles, and mild eosinophilia in some seminiferous tubules. Following cessation of treatment, some evidence of regeneration of spermatocytes was apparent beginning at 2 weeks. Decreased LDH-X isoenzyme activity was noted in testis homogenates.
