    The advantages and limitations of transdermal drug delivery were reviewed with regard to the possibilities of the method for systemic therapy. Advantages of transdermal drug systems included precise control over the plasma drug concentration, decreased side effects, elimination of first bypass metabolism, minimal inconvenience and anxiety for the patient, reduction of the frequency of dosing, enhanced patient compliance, and easy and quick termination of treatment if necessary. The primary limitations of the transdermal method were related to the barrier function of the skin, percutaneous metabolic effects, development of tolerance, and irritant or allergic reactions. Several types of transdermal therapeutic systems were described including membrane moderated systems, matrix diffusion controlled systems, and microsealed systems. The biological, physicochemical, and pharmacokinetic drug selection criteria for transdermal delivery were presented and assessed in relation to those drugs currently approved by the Food and Drug Administration for transdermal systems including estradiol, scopolamine, clonidine, and nitroglycerin; and the extension of the therapeutic potential of transdermal drug delivery with the use of penetration enhancers was reviewed.
