Neural specificity of acrylamide action upon enzymes associated with oxidative energy-producing pathways. I. Histochemical analysis of NADH-tetrazolium reductase activity.

    The effect of acrylamide (79061) (AA) and methylene-bisacrylamide (110269) (MBAA) on NADH-tetrazolium-reductase (NADHTR) activity in neural and nonneural tissues was studied in rats. Male adult Sprague-Dawley-rats were injected intraperitoneally with 50mg/kg AA or 104.8mg/kg MBAA. The control group received an equal volume of saline. One hour after injection dorsal root ganglion, cerebellar vermis, kidney, liver and stomach were dissected, frozen and sectioned. Another group of animals were administered AA, MBAA or saline 72 hours after ligation of the sciatic nerve. These animals were killed 1 hour later and the swollen nerve proximal to the ligation was prepared for examination. All tissue sections were incubated for demonstration of NADHTR activity, which was measured with a microdensitometer linked to a photomultiplier tube on a universal microscope. The order of decreased NADHTR activity for tissues was hepatocytes, parietal cells, kidney tubules, dorsal root ganglion, cerebellum and axons. Averaging a large number of randomly selected cells from each sample resulted in low variation of sample means. AA decreased the NADHTR activity in neural tissues without significantly decreasing enzyme activity in the nonneural tissues. The NADHTR activity was not significantly decreased by AA in the stomach, kidney or liver. MBAA was without a significant effect on NADHTR activity within any of the tissues with the exception of the Purkinje neurons. The author concludes the data support the hypothesis that specific inhibition of neural NADHTR activity by AA is the primary cause of neuropathy. [Description provided by NIOSH]
