Description:The effects of acute oral administration of lithium (7439932) compounds on multiple schedule performance were studied in rats. Adult male Sprague-Dawley-rats were administered lithium-chloride (7447418) or lithium-carbonate (554132) orally in doses of 100, 200, or 300 milligrams per kilogram (mg/kg). Bar pressing performance on a multiple fixed ratio 20/fixed interval 5 minute schedule was measured daily starting 3 days prior to dosing and continuing to 4 days after dosing. The 200 and 300mg/kg doses of lithium-chloride and all doses of lithium-carbonate caused significant decreases in performance. The 100 and 200mg/kg doses of lithium-carbonate had maximum effects on the day of administration, while the 300mg/kg dose had its maximum effect on the following day. Lithium-chloride showed a similar delay in peak effect for the 200mg/kg and 300mg/kg doses, although the effects were not as pronounced as with lithium- carbonate. The authors conclude that oral administration of lithium- chloride or lithium-carbonate affects operant behavior. Data is consistent with previously published results that show that peak brain concentrations are not reached until 24 hours after dosing with high lithium concentrations. The behavioral effects parallel brain lithium concentrations, but not serum lithium concentrations. [Description provided by NIOSH]
