Description:The pharmacologic properties of water soluble extracts of paper dust (PDE) were investigated using isolated guinea-pig trachea. Dose response relationships between PDEs and contraction of guinea-pig tracheal smooth muscle were established. Paper dust was obtained from separate sites in a paper recycling mill, both early and later in the process. Pretreatment with mediator blockers, arachidonic- acid metabolite inhibitors, and an intracellular calcium antagonist were examined and compared to findings with similar experiments using cotton bract extracts. The effect of pretreatment with capsaicin was also assessed. Contraction induced by paper dust extract collected early in the process was partially inhibited by all the mediator inhibiting agents as well as the receptor blocking agents with the exception of LY-171883. Similar findings were noted for paper dust extract collected late in the process, except that pyrilamine and indomethacin failed to block the bronchoconstrictor effect. Pretreatment with capsaicin enhanced the effects of both early and late paper dust extract. The authors suggest that water soluble extracts from the paper recycling industry cause airway smooth muscle constriction via mediator release. [Description provided by NIOSH]
