Description:F344/N-rats and B6C3F1-mice of both sexes were exposed whole body to vapors of N,N-dimethylformamide (68122) (DMF) at concentrations of 50, 100, 200, 400, or 800 parts per million (ppm) for 6 hours a day, 5 days a week for 13 weeks. Body weight gains were reduced by 50 to 65% in rats exposed at 800ppm and to a lesser extent in those exposed to 400ppm. All exposure concentrations produced an increase in serum cholesterol levels. Liver weights were increased in male rats exposed at 100ppm or higher and in all exposed female rats. Rats of both sexes exposed at 400 and 800ppm demonstrated minimal to moderate centrilobular hepatocellular necrosis. Body weight gains in mice were 30% less than in controls in females exposed at 800ppm. Genetic toxicity studies conducted in Salmonella-typhimurium with and without S9 metabolic activation indicated that DMF was not mutagenic. Germ cell mutations were not inducted in male Drosophila- melanogaster. No induction of sister chromatid exchanges or chromosomal aberrations were seen in-vitro in Chinese-hamster-ovary cells. The author concludes that the rats were more severely affected than mice. For both sexes of rats, the no observed adverse effect level was 200ppm. [Description provided by NIOSH]
