Description:The effects of pretreatment with tetrandrine on the pulmonary responses of the guinea-pig to inhalation of cotton dust were studied. English-short-hair-guinea-pigs were pretreated for 5 days with tetrandrine or saline orally; after pretreatment, guinea-pigs were exposed to cotton dust aerosols for 4 hours at a mass concentration of about 17mg/m3. In unexposed animals, oral administration of tetrandrine at 33 micrograms/gram body weight for 5 days had little effect on the number of lavagable macrophages of leukocytes. However, in these unexposed guinea-pigs, tetrandrine did inhibit breathing rate and the ability of macrophages to secrete superoxide anion in response to zymosan particles. In contrast, with cotton dust exposure, tetrandrine pretreatment accentuated cotton dust induced increases in breathing rate, number of lavagable leukocytes, and secretory activity of alveolar macrophages. The authors conclude that tetrandrine was apparently not useful in the prevention of pulmonary response of guinea-pigs to inhalation of cotton dust. [Description provided by NIOSH]
