Carcinogenesis of Roofing Asphalts, Pitch and Simulated Sunlight.

  An 18 month experiment was conducted to determine the tumorigenicity of volatile components of roofing asphalts to nonpigmented Swiss-CD- 1-mice and pigmented C3H/HeJ-mice. Four initial roofing materials were tested including two types of asphalt (8052424) and two types of coal-tar-pitch (65996932). Generation temperature was either 232 or 316 degrees-C. The other variable in the study was the presence of absence of sunlight. Fifty microliters of each test material was applied twice a week to each mouse using disposable tip automatic pipettes. Gross pathological examination of all necropsied animals revealed significant abnormalities in the liver, spleen and lung of the treated animals compared to controls. The generation of tumors in animals exposed to asphalt fumes appeared to be affected significantly by changes in temperature with the 316 degree setting being more active in tumor promotion. Animals exposed to coal-tar- pitch fumes did not demonstrate so significant a temperature effect. Few differences were noted in the same animal strain resulting from exposure to condensates of the two different types of asphalt or coal-tar-pitch materials generated at the same temperatures. The male-C3H mouse was far more sensitive to the tumorigenic activity of the asphalts than was the CD-1-mouse. The appearance of tumors was inhibited somewhat by the use of simulated sunlight.
