Description:Validation studies of the chromogenic Limulus amebocyte lysate (LAL) test for quantitating endotoxin activity in environmental dusts were described. The LAL test was used to determine the endotoxin content of a commercial lipopolysaccharide (LPS) derived from Escherichia- coli strain 0127:B8 dissolved in sterile nonpyrogenic water or saline. The LPS concentration was 500 nanograms per milliliter (ng/ml). The endotoxin concentrations in the aqueous and saline solutions were 3934.72 and 2743.66 endotoxin units per milliliter (EU/ml), respectively. The LAL test was used to determine the endotoxin content of eight commercial LPSs derived from E-coli, Salmonella-typhimurium, S-typhosa, and S-minnesota. The endotoxin concentrations varied widely, ranging from 0.31EU/ml in the S- minnesota LPS to 3934.72EU/ml in the E-coli 0127:B8 LPS. To determine the extent of endotoxin contamination, the LAL test was applied to solvents used for extracting LPS samples. Human and chicken albumin contained measurable amounts of endotoxin. Samples of spring wheat and oat dust were extracted with 10ml water, tween- 20, tween-80, triton-X-100, saponin, and human, bovine, and chicken albumin and assayed for their endotoxin content using the LAL test. The endotoxin content of the dusts varied widely, depending on which extraction solution was used. The authors conclude that the chromogenic LAL test can be used to determine the endotoxin content of environmental dusts. Using extraction solutions other than water or those specified by the test kit manufacturer may produce erroneous results. [Description provided by NIOSH]
