Description:Whole embryo and limb in-vitro teratogenic assay systems are discussed. Factors influencing in-vitro test results are considered along with various types of assay systems. Procedures used in tests with embryo limb buds are described and tabulated results on tests with various teratogenic agents are provided. The effects of activating agents on teratogenic potency are evaluated. The author concludes that limb bud assays are less expensive and less time consuming than in-vivo tests. The buds can be exposed during organogenesis and can be maintained for assessment of differentiated structure and function. The major limitation involves the inability to assess the response of the intact embryo, which may differ from that of the isolated buds. [Description provided by NIOSH]
