Description:Laboratory procedures for assessing human semen quality in epidemiological studies were discussed. The discussion was a summary of a meeting of andrologists who were currently using computer assisted semen analysis (CASA) techniques and endpoints in field and clinical studies in the spring of 1990. Guidelines for obtaining semen samples from donors were discussed. Factors that could affect semen quality such as abstinence time before obtaining a sample, use of masturbatory aids, and sample age were considered. Recommendations for handling semen samples were reviewed. Measuring sperm motility was discussed. Procedures for diluting semen samples were described. It was noted that establishing a standardized method for diluting semen samples is important because the existing CASA systems cannot accurately determine curvilinear velocity, straight line velocity, amplitude of lateral head displacement, or degree of linearity in samples containing sperm concentrations greater than 40 million per milliliter. Equipment recommendations for CASA systems were discussed. Proper machine settings for obtaining optimum semen data with the existing CASA systems were described. [Description provided by NIOSH]
