Description:Monoclonal antibodies were developed for the detection of benzo(a)pyrene (50328) (BaP) and its metabolites in urine. Spleen cells from Balb/C-mice immunized with BaP linked to bovine serum albumin were isolated and fused with myeloma cell line P3x63- AG.8.653. Antibodies were purified and named 4D5 and 10C10. An immunoaffinity column of antibody 4D5 was made to concentrate BaP from urine. A competitive enzyme linked immunosorbent assay (ELISA) was used for quantitative measurement of BaP and metabolites. At four picomole BaP, there was a 50% inhibition with both antibodies. The antibodies also cross reacted with the metabolites of BaP and with other polyaromatic hydrocarbons (PAH). Mice were injected with tritium labeled BaP and urine was collected to determine if these antibodies could be used with biological samples. Analysis with 4D5 gave 50% inhibition at 12 to 15 picomole BaP metabolite. The authors conclude that this assay is useful as a general indicator of exposure though the antibodies cross react with other PAH so quantitation is difficult. [Description provided by NIOSH]
