Enhanced IL-1 beta and tumor necrosis factor-alpha release and messenger RNA expression in macrophages from idiopathic pulmonary fibrosis or after asbestos exposure

Description:Bronchoalveolar lavage (BAL) was performed on patients with idiopathic pulmonary fibrosis (IPF) and on individuals who had asbestos (1332214) exposure with radiographic evidence of parenchymal fibrosis to obtain alveolar macrophages (AM) for analysis of IL-1beta and TNF-alpha protein release and gene expression. BAL recovered significantly more cells in asbestos exposed and IPF patients than in normal controls. Both patient groups released significantly more TNF-alpha than controls. Only the asbestos exposed subjects released significantly more IL-1beta than controls. A significant correlation was found between the number of neutrophils per milliliter of BAL fluid recovered and the spontaneous release of TNF-alpha for both groups combined. AM lavaged from patients expressed increased mRNA levels for both IL- 1beta and TNF-alpha. The increased production of IL-1beta and TNF- alpha was caused by enhanced expression of the mRNA for the cytokines. The authors note that the cytokines have the potential to influence the remodeling and fibrosis observed in the lower respiratory tract in IPF and asbestosis. [Description provided by NIOSH]
