Endotoxin and complement activation in a analysis of environmental dusts from a horse barn

  Endotoxins were quantified in bulk dusts and material obtained from a horse barn that was associated with reports of respiratory problems in workers. The activity of a sample against human serum complement in-vitro was used to quantify its inflammatory potential. Bulk dust samples were obtained from several areas within the horse barn, including the loft area, delivery chute and exterior of the feed bin, and bulk bedding material. Samples were extracted with water, and extracts were analyzed for endotoxin content and activity against human complement in-vitro. Every sample contained quantifiable contamination with endotoxins ranging from 58.20 to 119.14 endotoxin units/milligram (EU/mg). Endotoxin in samples of bedding material ranged from 195.31 to 30273.44EU/mg. The extracts prepared showed activity against human complement in a dose dependent manner. The bedding material was much more active. The findings suggested that gram negative bacterial endotoxins were present in all bulk dusts and materials tested and that the levels of contamination were similar to those in other agricultural dusts.
