Effects of ammonium metavanadate treatment upon macrophage glutathione redox cycle activity, superoxide production, and intracellular glutathione status

Description:B6C3F1-mice were administered ammonium-metavanadate (7803556) at 10mg/kg, or 2.5mg/kg as vanadium (7440622) intraperitoneally every 3 days for 6 weeks, and the effects on three enzymes involved in energy production for phagocytosis and for the removal of peroxides, as well as the production of superoxide anion were studied. An attempt was made to determine whether vanadate pretreatment could contribute to the observed depressed phagocyte responses and Listeria killing by inhibiting enzymes related to the respiratory burst in peritoneal macrophages (PEM). These enzymes included glucose-6-phosphate-dehydrogenase, glutathione-reductase and glutathione-peroxidase. The results clearly showed that intraperitoneal exposure of mice to vanadium for 6 weeks could affect the three cytoplasmic enzymes which are crucial for PEM function, as well as the level of substrate required for their proper activity. The authors suggest that their 6 week exposure period may serve as a model for analyzing the effects of vanadium on macrophage function after long term exposure. [Description provided by NIOSH]
