Description:A sampling and analytical procedure was developed for measuring urine concentrations of benzidine (92875), ortho-dianisidine (119904), ortho-tolidine (119937), and 4,4'-methylenedianiline (101779) for use in industrial hygiene investigations. After hydrolysis of urine metabolites, the free diamines were isolated using a C18 solid sorbent. The free diamines were eluted, concentrated, and injected into a high performance liquid chromatography system. Using a C18 column with a mobile phase of acetonitrile and water, separation was achieved. By monitoring the ultraviolet absorbance at 280 or 245 nanometers and the electrochemical response, identification and quantitation were achieved. The limit of detection for these aromatic diamines was less than 2 micrograms/liter using ultraviolet detection while the limit of quantitation was less than 6 micrograms/liter. For electrochemical detection the limit of detection was less than 0.3 micrograms/liter and the limit of quantitation was less than 0.9 micrograms/liter. Recoveries ranged from 87 to 102% at 2, 10, and 20 micrograms/liter levels. [Description provided by NIOSH]
