Description:Methods for identification and analysis of particulates in biological tissue are viewed. Types of particles commonly found in tissue are listed, including mineral deposits such as calcium salts, uric-acid, or urate crystals; exogenous particles from external sources such as sodium or potassium oxalate, antacid agents and demulcents, pigments, and respirable dusts; and endogenous particles such as melanin, hematin, and hemosiderin. Various techniques of studying particles in situ, including light microscope, transmission electron microscope, and scanning electron microscope (SEM) and their various imaging modes are described. Identification of inorganic particles by scanning electron microscopy and analysis by backscattered electron imaging (BSE) are discussed. Staining and autoradiographic tagging of organic particles and the disadvantages of using BSE techniques are presented. Other detection methods used with SEM are discussed. Techniques for in situ tissue removal and analysis are described. Wet digestion, ashing under high and low temperatures, and ion etching procedures also are described. The results of a comparative test on lung tissue analyzed by wet digestion, ion itching, and low temperatures are evaluated. [Description provided by NIOSH]
