    The action of 2,4-toluenediamine (95807) (TDA) on spermatogenic tissues, its effects on androgen binding protein (rABP) production, on ultrastructural changes in seminiferous tubules, and on testicular weights and morphology were investigated in rats. Male Sprague-Dawley-rats, were placed on diets of ground chow containing conducted. In the first experiment, serum, medium of seminiferous tubule cultures, and testes and epididymal cytosol were analyzed for rABP content. Experiment two looked at testicular and epididymal sperm concentrations during 4 to 10 weeks of feeding with 0.03 percent TDA. Experiment three tested the responsiveness of tubular physiology to 0.06 percent TDA fed for 1 to 3 weeks. The results of experiment one showed a seven to nine fold increase in rABP content in testicular cytosol or in media of cultured seminiferous tubules. Examination of testicular tissue revealed variable degenerative changes in Sertoli cells as compared with controls, including cytoplasmic swelling, membrane disruption, and vacuolization. Experiment two showed a doubling of testes/body weight ratios and a 2.5 to 2.9 fold increase in seminiferous tubule fluid volume. Experiment three resulted in a 25 percent decrease in epididymal sperm content, reduced epididymal weight, and minor changes of structure of Sertoli cells. Results indicated that TDA is toxic to testicular spermatogenesis within 1 week of feeding, and that at the third week of TDA feeding, the toxicity is expressed in further alterations in Sertoli cell structure and reduction in epididymal sperm reserves. The authors conclude that the Sertoli cell is a locus of action in the spermatogenic disruption of the TDA treated rat. [Description provided by NIOSH]
