Description:The use of rabbits to study byssinosis was evaluated. New-Zealand-white-rabbits were anesthetized and intubated to study lung function including transpulmonary pressure. Resistance and compliance values and the percent change from baseline values were calculated as a function of time after inhalation of 10 milligrams per milliliter (ml) histamine or 1 gram of cotton dust extract per 25ml water. Twelve of 14 rabbits were characterized as histamine responders with an increase of 25 percent above baseline values for resistance and compliance. Three of these were immediate maximal responders while nine had delayed maximal responses at 5 or 10 minutes. Four of the 12 were mild responders, 2 were moderate and 6 were severe responders. Ten of 14 rabbits were clinically significant responders to cotton dust extract. Response was immediate in all but two of them. Response in one was delayed and mild, but of the others, three were mild; one, moderate; and six, severe responders. The non histamine responders also did not react to cotton dust extract. Two non responders to cotton did not react to histamine. All cotton dust responders reacted to histamine. The authors conclude that pulmonary response after inhalation of cotton dust extract can be measured in rabbits as a model for byssinosis in humans. [Description provided by NIOSH]
