An evaluation of the copulatory, endocrinologic, and spermatotoxic effects of carbon disulfide in the rat.

Description:The effects of carbon-disulfide (75150) (CS2) on a reproductive function were examined in male Long-Evans-Hooded-rats. Rats were exposed to 607 parts per million CS2 in a exposure chamber for 6 hours per day, 5 days per week, for 10 weeks. Before exposure, baseline values were established for copulation behavior, semen parameters, semen plug weights, and hormone concentrations. Animals were evaluated for these parameters at weeks 1, 4, 7, and 10 of exposure. Blood samples were taken and assayed for follicle stimulating hormone (FSH), testosterone, and luetinizing hormone (LH). Mount and ejaculation latencies initially decreased in both exposed and sham exposed controls. However, they were of greater magnitude in CS2 exposed animals by week 4, and were significantly different by week 10 of exposure. No effects in number of mounts or ejaculations were seen in exposed rats; the same numbers were performed within a shorter time frame. Significant depressions in sperm counts were seen at weeks 7 and 10 of exposure. No treatment related effects were seen in analysis of testosterone, FSH, or LH. Prostate weights in CS2 exposed rats were significantly less than in controls. No treatment related effects were seen in sperm counts from the epididymis. There were no histological changes in epididymis or testis. The authors conclude that CS2 had no apparent effect on the hypothalamic pituitary testicular axis. Decreases in prostatic weight and seminal plug weights suggest a target tissue insensitivity to androgen action. [Description provided by NIOSH]
