Use of hexyl isocyanate antigen to detect antibodies to hexamethylene diisocyanate (HDI) in sensitized guinea pigs and in a sensitized worker.

Description:The production and specificity of antibodies produced in guinea-pigs following intradermal injection of Hexamethylene-diisocyanate (822060) (HDI) and hexyl-isocyanate (HMI) were studied. Antigens were also tested for ability to detect antibodies in a worker with clinical HDI sensitivity. Groups of female English-smooth-haired-guinea-pigs were sensitized by intradermal injection of 50 microliters HDI or 50 microliters HMI. A serological assay for HDI sensitivity was performed. Human serum from a car painter sensitized to HDI was assayed to detect specific IgE antibodies as well as the ability of various isocyanate conjugates to inhibit radioallergosorbent tests (RAST). Guinea-pigs injected with HDI produced antibodies which precipitated with HDI guinea-pig serum albumin (GSA). Guinea-pigs injected with HMI produced antibodies which precipitated only with HMI-GSA. Results of passive cutaneous anaphylaxis (PCA) produced titers with high concentrations of HDI-GSA. The PCA technique used to evaluate specificity of antihexyl isocyanate antibodies produced the greatest reactions with HMI-GSA and n-butyl-isocyanate GSA. Antibodies to HMI differed from anti HDI antibodies in their lack of requirement for GSA determinants. HDI-GSA was the more potent inhibitor of the reaction to determine specificity of antibodies induced by HDI. Serum from the sensitized patient found 7.2 percent of the added radioactivity in the HDI human serum albumin (HSA) RAST. When 100 micrograms HDI-HSA was incorporated into the assay buffer, binding was reduced to 1.4 percent. The patient's serum was also positive in the RAST assay. The authors conclude that HMI and HDI are immunogenic in guinea-pigs. HMI induced predominantly hapten specific antibodies whereas HDI induced antibodies specific for aliphatic isocyanate-GSA determinants. [Description provided by NIOSH]
