Description:Multicellular in-vivo sister chromatid exchanges (SCE) induced by urethane (51796) were studies in male BDF-1-mice. Partially hepatectomized and intact mice were exposed for 4 hours on day 5 post hepatectomy to 0.035, 0.11, 0.22, 0.475, 0.90, or 2.06 milligrams per liter (mg/l) urethane (51796). Mice were also given intravenous infusions for 4 hours or a single dose intraperitoneally (ip) of 193mg per kilogram urethane prior to 5-bromo-2'-deoxyuridine (59143) (BrdU) incorporation testing. On day 6 post hepatectomy, mice were given an ip injection of 100 micrograms of colchicine and were sacrificed 4 hours later. Clear dose relationships were apparent in all cell types in both hepatectomized and intact mice. No significant increase in SCE frequencies were observed at 0.035mg/l, but at doses of 0.1mg/l and greater, induced SCE frequencies were linearly related to urethane concentrations. No significant differences in intact and hepatectomized mice were noted. Bone marrow response was significantly lower than that of alveolar macrophages. Inhalation and intravenous infusions of the same total dose of urethane produced comparable SCE responses, but a single ip injection produced significantly higher SCE frequencies in bone marrow and alveolar macrophages.
