Determination of the reproductive effects in mice of nine selected chemicals.

Description:The reproductive effects of sodium-selenite (10102188) (SS), ethylene-thiourea (96457) (ETU), diethylene-glycol-monomethyl-ether (111773) (DEME), ethylene-glycol-monomethyl-ether (109864) (EME), ethylene-glycol-monoethyl-ether (110805) (EMEE), ethylene-glycol- monobutyl-ether (111762) (EBE), toluene (108883), 2,4-dinitrotoluene (121142) (2,4DN), and 2,4-diaminotoluene (95807) (2,4DA) were examined in mice. Female CD1-mice were given 10 milliliters per kilogram oral doses of each compound dissolved in an appropriate solvent at various concentrations to determine the maximum tolerated dose (MTD). Pregnant mice were administered the compounds orally for 8 days, the concentrations of each compound being based on the MTDs. Animals were observed for clinical symptoms and any dead mice were necropsied. Litter size, number of live pups, and litter weights were tabulated and statistically analyzed. The MTDs of SS and toluene caused no significant reproductive toxicity. ETU caused a significantly reduced number of live pups per litter. EBE and 2,4DN resulted in a large number of resorptions and maternal defects; however, the viability of the litters was not significantly affected. DEME and 2,4DA caused a large number of resorptions and maternal deaths as well as a large number of stillbirths. EME and EMEE caused extreme reproductive toxicity. No viable litters were observed.
