N-hexane-"free" hexane mixture fails to produce nervous system damage.

Description:The effects of chronic inhalation of a mixture of hexane isomers virtually free of n-hexane (110543) on a rat species known to develop n-hexane neuropathy were studied. Groups of six animals were housed in four inhalation chambers and exposed to 1,475 milligrams per cubic meter (mg/cum) of purified methyl-ethyl-ketone (78933) (MEK) vapors, a 1,750mg/cum mixture of hexane isomers largely free of normal hexane, 400mg/cum of methyl normal butyl- ketone (591786) (MnBk), or to hydrocarbon free air. Two animals chosen randomly from each test group were submitted to detailed neuropathological examination following 2, 4, and 6 months of exposure. During the exposure period, none of the test animals displayed neurological impairment, or any other clinical disorder. No significant clinical or histopathological differences were observed between control animals and animals exposed to MEK or to the mixture of n-hexane free hexane isomers. Beginning nerve fiber damage appeared at 4 months in animals exposed to MnBk, and was substantially advanced by 6 months. The authors conclude that hexane isomers virtually free of n-hexane are devoid of neurotoxic properties when tested at a total concentration of 1,750mg/cum, and recommend further study in experimental species to determine the possibility of human neuropathy caused by exposure to subthreshold concentrations of n-hexane.
