Description:The NIOSH validated method for the determination of p-nitroaniline (100016) (PNA) is described. Ninety liters of air (flow rate at 1.5 liters per minute) are drawn through a 37 millimeter diameter mixed cellulose ester membrane filter to trap PNA. The filter is transferred to a 2 ounce sample jar and extracted with 5 milliliters of isopropanol. A 20 microliter aliquot is injected into a high pressure liquid chromatograph equipped with a variable ultraviolet detector set at 375 nanometers. The PNA concentration in the sample is determined by reference to a calibration curve obtained by analyzing standards having concentrations of 0.1 to 3 times the OSHA standard of 6 milligrams per cubic meter (mg/m3). PNA samples are stored for 7 days and analyzed after day 1 or day 7. The average recovery is 96.8 and 98.6 percent for the 1 and 7 day old samples, respectively. The useable range of the method is 3.9 to 12.9mg/m3 PNA with a standard deviation of 0.326mg/m3. The detection limit of the method is estimated to be on the order of 10 micrograms per milliliter.
