  In this review of the use of monoclonal antibodies for the detection of aberrations in the expression and structure of specific genes, the authors discuss briefly the usefulness and criteria for antibodies to be used in biological monitoring attempts. The authors express concern that standards need to be set as additional resources are being committed to this approach. The specificity of antibodies has apparently been assumed frequently on the basis of a visual assessment of the pattern of labeling and such evidence has not been shown to be unequivocal. They note that the most obvious manner to demonstrate the specificity of an antibody is through demonstrating competition with the authentic protein. They propose that any work purporting to demonstrate the use of antibodies to oncogene proteins for monitoring purposes be considered as preliminary until the specificity to the antibody is able to be established by competition or by more complex biochemical isolation of protein binding to the antibody in immunochromatography. A quantitative relationship must also be demonstrated between the amount of antibody bound and the signal observed. Unless all of the immunological reagents are at saturating conditions, such a quantitative relationship can not be established. [Description provided by NIOSH]
