Concentration dependence of pulmonary responses of the guinea pig animal model following inhalation of endotoxin derived from Enterobacter agglomerans.

Description:Studies were conducted in guinea-pigs to determine whether endotoxin alone in concentrations similar to those found in aerosolized cotton dust would cause pulmonary responses similar to those observed in past animal studies using cotton dust. English-short-hair-guinea- pigs were exposed to aerosols of nebulized endotoxin extracted from Enterobacter-agglomerans for 3 hours; concentrations of endotoxin ranging from 0.1 micrograms/milliliter to 3.3mg/ml. At 18 hours following exposure, breathing rate, total lavagable cells, lymphocytes and red blood cells increased in a dose dependent manner, peaking at 1.0mg/ml. Granulocytes were increased by 74 fold at 1.0mg/ml before declining to a 34 fold increase. Alveolar macrophages initially declined by 27% at 0.1 micrograms/milliliter endotoxin prior to increasing by 57%. Zymosan stimulated superoxide release increased 102% at 3.3mg/ml. The authors conclude that the inhalation of E-agglomerans endotoxin results in pulmonary responses which are qualitatively similar to those seen with cotton dust. [Description provided by NIOSH]
