Description:A review of stimulus secretion coupling in phagocytic cells, based on a general scheme for stimulus secretion systems, was presented. An overview of stimulus secretion coupling in other cell systems was presented and a general scheme was proposed. One problem in studying stimulus response coupling in phagocytic cells is the fact that data are derived from four different cell types: peripheral granulocytes, peritoneal neutrophils, peripheral monocytes, and alveolar macrophages. Binding of stimulants to membrane receptors and characteristics of these receptors were described. Stimulant induced changes in transmembrane potential measured by four techniques were considered. Strong, although not conclusive, evidence was presented for the role of depolarization of phagocyte membranes in stimulus secretion coupling. Stimulant induced changes in membrane transport properties were addressed. Mobilization of intracellular calcium as a factor for secretory activity of phagocytes was discussed. The role of calmodulin in the stimulation response scheme of phagocytes was considered. Involvement of cyclic- adenosine-monophosphate in the stimulus secretion cycle was addressed. Stimulant induced protein phosphorylation was considered. Stimulation of lipid metabolism and its role in phagocytosis were described. Stimulant induced changes in cytoskeletal components were discussed. A working hypothesis for the stimulus response coupling cascade of phagocytic cells was illustrated.
