Description:In order to determine whether heterogeneity exists between pulmonary macrophages collected from lavage fluid, an investigation was performed to characterize the cells collected from different lung washes. Cells were isolated from male pathogen free Sprague-Dawley- rats. The morphology of cells obtained by pulmonary lavage varied with the lavage number. In general, cells from lavage number one were larger. The cytoplasm of the cells from the first lavage appeared less electron dense and contained more vacuoles. A large amount of extracellular debris was found in the first lavage which was not present in later ones. The purity of the cells, in terms of pulmonary macrophages per total number of cells, was similar for each lavage fraction. The yield of pulmonary macrophages was highest in lavage number two. No differences were noted in either unstimulated or stimulated oxygen consumption of cells from different lavages. However, cells from different lavages did vary with respect to their capacity to release reactive oxygen species in response to stimulation with zymosan. Zymosan stimulated release of reactive compounds was lowest with cells removed during early lavages. Cells from the first lavage produced less superoxide anion in response to stimulation with zymosan particles. However, the unstimulated rate of superoxide anion production did not vary between lavages. In response to zymosan stimulation, the cells from lavage number one released less hydrogen-peroxide than other lavages, while unstimulated activity did not vary between cells from different lavages. Pulmonary macrophages removed early in the lavage process released lesser amounts of reactive oxygen species upon stimulation with zymosan particles than did cells removed later in the lavage process.
