Potentiating and inhibitory effects of periodate-oxidized ATP analogs on contractions of vas deferens to ATP

Description:A study was undertaken to compare the periodate oxidized analogs of ADP, AMP, and adenosine against periodate oxidized ATP for their ability to initiate contractile responses, to potentiate the first phase of response to ATP, and to inhibit the second phase response. Vasa deferentia were isolated from English-short-hair-guinea-pigs and attached to a holder in an organ batch containing 1 milliliter (ml) of modified Krebs/Henseleit solution. Isometric contractions were measured by attaching the vasa deferentia to force displacement transducers. Tissues were incubated for 5 minute (min) intervals with periodate oxidized ATP, ADP, AMP, and adenosine. ATP concentration curves were determined using a 13min addition cycle. Vas deferens contraction was initiated by periodate oxidized ATP and ADP. The first phase of contraction was potentiated following preparation incubation with periodate oxidized ATP, ADP, AMP, and adenosine. The second phase of contraction was inhibited by periodate oxidized ATP, periodate oxidized ADP and periodate oxidized AMP. The authors conclude that the periodate oxidized analogs of ADP, AMP, adenosine, and ATP potentiate the first phase of contraction to ATP in guinea-pig isolated vas deferens, and that all of these analogs except for periodate oxidized adenosine inhibit the second phase.
