Influence of the dose levels of cocarcinogen ferric oxide on the metabolism of benzo(a)pyrene by pulmonary alveolar macrophages in suspension culture

Description:The effects of the phagocytosis of ferric-oxide (1309371) (FeO) by alveolar macrophages on the metabolism of benzo(a)pyrene (50328) (BaP) and on the production of reactive oxygen species were studied in-vitro. Alveolar macrophages were harvested by tracheal lavage from Syrian-Golden-hamsters, cultured with dimethylsulfoxide and cytochrome-c, and exposed to radiolabeled BaP that had previously been adsorbed onto FeO. A decrease in cell viability was seen that was dependent upon the dose of BaP coated particles present in the culture media. A significant increase in the amount of reduced cytochrome-c was seen up to 6 hours after the addition of BaP coated FeO or FeO alone compared with the amount reduced in untreated cells or cells incubated with BaP alone. The amount of reduced cytochrome- c plateaued in all preparations after 6 hours. The highest concentration of phagocytosed BaP was seen in cultures exposed to BaP coated FeO at 3 hours. The phagocytosed BaP was metabolized by the macrophages and the total concentration of metabolites produced appeared to be dependent on the dose of FeO.
